Ie: I have noticed my targets are usually restocking toys on mon/tues am. I am sure all the stores are different, but perhaps if we share what we have noticed or know, we can compile some consistencies.

I think it really depends on individual markets, but at my Target they get trucks in overnight before hours on Monday, Wednesday, and Friday. So those three mornings — as well as Saturday morning — are always good for new pickin's.
My Kmart gets trucks on Thursday and/or Friday, so I can usually find signs of life between Thursday evening and Saturday morning.
I have no idea about my two Wal-Marts, as they are on Toy Manager #64 since I started collecting 5+ years ago, but judging by their inventory, nothing has arrived since the 2nd wave of stuff hit the endcaps (cubes at other peoples' Wal-Marts).
TJ Maxx gets trucks on Monday, Wednesday, Thursday.
Gerbes (Krogers) gets trucks on Monday and Friday, but since the initial launch of Cars 2 stuff, they seem to be pretty much out of the Cars market.
Kohls hasn't had a truck since 1983.
KB Toys is of course long since closed and out of business, but they used to call me every Monday with new stock. Oh how I miss them!
Sadly, there is no Toys R Us in town.

Has anybody noticed the new cube at your KMart? I have not. I've checked 4 KMarts in the past 2-3 days.

Due to a lack of sufficient lighting in most Kmarts, the vast majority of store managers misread "CARS Super Cubes" in the corporate fax as "C&H Sugar Cubes". So get ready to brew LOTS of coffee and tea this Fall!
